To investigate the mechanism of inhibition of RBF oncholesterol synthesis, the effect of RBF on the incorporation intocholesterol of radiolabeled acetate and radiolabeled mevalonate inrat hepatoma cell culture was measured. The results of thisexperiment suggested that the inhibition of cholesterol synthesisappears to be mediated primarily at the level of HMG-CoA reductase,since inhibition of the incorporation of radiolabeled acetate wasmarkedly higher than that of radiolabeled mevalonate, the productof HMG-CoA reductase.
