Synthesis and Properties of N,N-Dimethyl-O ..

The product is dissolved in sodium chloride containing aqueous solutions and reprecipitated by ethanol, again isolated by centrifugation and thoroughly washed with ethanol and diethyl ether 22.Another method for quaternisation of chitosan is by way of reacting with betaine in the presence of the coupling reagent 2-ethoxy-1-ethoxycarbonyl-1, 2-dihydroquinoline (EEDQ) in aqueous media at pH 5.5 ± 0.5. This reaction results in preparation of N-(trimethylammonio) acetyl/chitosan chloride and its amphiphilic derivatives. The degree of quaternization increases with increasing EEDQ/chitosan ratio and is partly followed by N-ethoxy carbonylation. That side-product formation can be slowed down by increasing betaine/EEDQ ratio.23

The CTS-PVP film with iodine was prepared by chitosan(CTS) grafted polyvinyl pyrrolidone(PVP) in the protection of N2 with dimethyl 2, 2'-azobis(2-methylpropionate)(AIBME) as initiator followed by addition of a certain amounts of iodine in ethanol solution and a gelatin solution. The properties of the film were characterized by infrared(IR), scan electron microscopy(SEM), UV-visible spectroscopy(UV-Vis), differential scanning calorimetry(DSC), etc. The UV-Vis spectrum shows a strong absorption between 200~600 nm for the CTS-PVP film with iodine. The glass transition temperature at 319 ℃ indicates a good thermal stability. The antimicrobial test result shows that the inhibition zone diameters increase along with the increasing amounts of CTS-PVP-I2 complex liquid. The inhibition zone diameters against Staphylococcus aureus and Escherichia coli are in the range of 16~36 mm, when the added volume of CTS-PVP-I2 complex fluid is 10 mL or more, implying its high sensitivity and good antibacterial properties.

In this context, after the introduction of long chain quaternary ammonium groups, a chitosan derivative which was soluble in water was synthesized using CMCTS, decylalkyl dimethyl ammonium and epichlorohydrin. Compared with the main cosolvent currently used in sustained-release formulation, chitosan and its derivative have the advantage of being easily accessible, biologically secure and environmentfriendly. This polymer is suggested to be also biodegradable similar to chitosan because both of them have the same main body (-). The solubility determination experiment results showed that the solubility in water of this polymer was about 0.04 g·mL-1. In addition, this chitosan derivative could overcome some weakness of chitosan and possess some improved properties, such as being non-toxic and biocompatible. Generally, the sustained-release formulations of poorly soluble drugs have poor dissolution performance. Aspirin, a representative of those drugs was chosen to be a model drug. According to the characteristics of this biopolymer, the major objective of this work was to investigate chitosan derivative and its ability to increase aspirin release from sustained-release matrix tablets.

Although the (14)-anhydroglucosidic bond of chitin is also present incellulose the characteristic properties of chitin/chitosan are not shared bycellulose18. Chitin is highly hydrophobic and is insoluble in waterand most organic solvents. It is soluble in hexafluoroisopropanol, hexafluoroacetone, and chloroalcohols in conjunction with aqueous solutions ofmineral acids1 and dimethylacetamide (DMAc) containing 5 per centlithium chloride (LiCl)19. Recently the dissolution of chitosan in -methyl morpholine--oxide (NMMO)/H2O has been reported by Dutta 20,21. The hydrolysisof chitin with concentrated acids under drastic conditions produces relativelythe pure amino sugar, -glucosamine.

Chitosan has replaced the synthetic polymers inopthalmological applications. Chitosan possesses all the characteristicsrequired for an ideal contact lens; optical clarity, mechanical stability, sufficient optical correction, gas permeability, partially towards oxygen, wettability, and immunologically compatibility. Contact lenses are made from partiallydepolymerized and purified squid pen chitosan by spin casting technology, andthese contact lenses are clear, tough, and possess other required physicalproperties such as, modulus, tensile strength, tear strength, elongation, watercontent, and oxygen permeability. Antimicrobial and wound healing properties ofchitosan along with excellent film forming capability make chitosan suitablefor development of ocular bandage lens76.
